چکیده
Ab s t r a c t . The thermal degradation of Aspergillus oryzae α-amylase in the presence of sugars (sucrose, trehalose) and polyols (sorbitol, glycerol) as stabilization additives was investigated. The aim of the study was to test whether it is possible to estimate the heat stability of the enzyme based on the amount of hydroxyl groups provided in a buffer solution from different sources. The thermal inactivation experiments were performed at 68°C. Every additive tested showed a protective effect on the enzyme heat stability, the effect being strongly dependent on the added compound concentration. Among all the stabilizing compounds investigated, sucrose exhibited the greatest protective effect. The decimal reduction time of α-amylase activity increased 33.9 times when 420 mg/ml of sucrose was added to the environment. When the same concentration of trehalose was used, the D-value increased 6.4 times compared to the value in the buffer system. The number of hydroxyl groups provided in the enzyme solution could not be related to the D-values for the enzyme thermal inactivation, meaning that the enzyme heat stability was not dependent on the number of hydroxyl (OH) groups. Amylase production from Aspergillus oryzae LS1 by solid-state fermentation and its use for the hydrolysis of wheat flour
Nine Aspergillus and three of Trichoderma strains were grown on wheat bran (WB) medium under solid state fermentation (SSF) for amylase production. Aspergillus oryzae LS1 produced the highest level of the enzyme. The thermal stability profile of its crude enzyme revealed the half-life time of more than 2 h at 50 and 60ºC. The enzyme production was affected by strain type, incubation periods, le...
متن کاملGene silencing by RNA interference in the koji mold Aspergillus oryzae.
We found the orthologous genes required for RNA interference (RNAi) in the Aspergillus oryzae genome database, and constructed a set of tools for gene silencing using RNAi in A. oryzae. This system utilizes compatible restriction enzyme sites so that only a single target gene fragment is required to create the hairpin RNA cassette. For ease of handling, we also separated the construction of the...
متن کاملAlpha Amylase Production by Aspergillus Oryzae Using Solid State Fermentation
A fungal strain of Aspergillus oryzae was used for the production of alpha amylase by solid state fermentation from agro-industrial waste. Enzyme production was growth associated and maximum activity (8.23 U/ml) were obtained after 120h when incubated at 30°C on wheat bran with initial moisture content 60%; initial medium pH = 5 . Enzyme activity increased when the solid medium was supplemented...
متن کاملPresence of binding site for alpha-amylase and of masking protein for this site on mycelial cell wall of Aspergillus oryzae.
Mycelial cell wall of Aspergillus oryzae M-13 grown in an alpha-amylase-forming medium could not bind alpha-amylase (Taka-amylase A, EC 3.2.1.1). However, by treatment with 1.0 n NaOH at 100 C for 30 min, the wall gained the ability to bind alpha-amylase. This phenomenon was caused by removal of a factor (designated as masking factor) which masked the binding site for alpha-amylase. The masking...
متن کاملRapid induction of alpha-amylase by nongrowing mycelia of Aspergillus oryzae.
A rapid induction system for synthesis of alpha-amylase by the funga Aspergillus oryzae M-13 was established. The mycelia were prepared from 20-h cultures grown on a peptone-glycerol medium and starved for 5 h; maltose was the optimum inducer tested. During h 1 of induction, formation of both intra- and extracellular alpha-amylases occurred at an almost identical rate (70 to 80 microgram/g of c...
